چکیده

We have studied the effects of the local anesthetic lidocaine, and the general anesthetic halothane, on the function and oligomeric state of the Ca-ATPase in cardiac sarcoplasmic reticulum (SR). Oligomeric changes were detected by timeresolved phosphorescence anisotropy (TPA). Lidocaine inhibited and aggregated the Ca-ATPase in cardiac SR. Micromolar calcium or 0.5 M lithium chloride protected against lidocaine-induced inhibition, indicating that electrostatic interactions are essential to lidocaine inhibition of the Ca-ATPase. The phospholamban (PLB) antibody 2D12, which mimics PLB phosphorylation, had no effect on lidocaine inhibition of the Ca-ATPase in cardiac SR. Inhibition and aggregation of the Ca-ATPase in cardiac SR occurred at lower concentrations of lidocaine than necessary to inhibit and aggregate the Ca-ATPase in skeletal SR, suggesting that the cardiac isoform of the enzyme has a higher affinity for lidocaine. Halothane inhibited and aggregated the Ca-ATPase in cardiac SR. Both inhibition and aggregation of the Ca-ATPase by halothane were much greater in the presence of PLB antibody or when PLB was phosphorylated, indicating a protective effect of PLB on halothane-induced inhibition and aggregation. The effects of halothane on cardiac SR are opposite from the effects of halothane observed in skeletal SR, where halothane activates and dissociates the Ca-ATPase. These results underscore the crucial role of protein-protein interactions on Ca-ATPase regulation and anesthetic perturbation of cardiac SR.

Localization of Ca " + Mgt + - ATPase of the Sarcoplasmic Reticulum in Adult Rat Papillary Muscle ANNELISE O . JORGENSEN , AMY C

Localization of the Ca + Mgt+-ATPase of the sarcoplasmic reticulum in rat papillary muscle was determined by indirect immunofluorescence and immunoferritin labeling of cryostat and ultracryotomy sections, respectively . The Ca + Mgt+ -ATPase was found to be rather uniformly distributed in the free sarcoplasmic reticulum membrane but to be absent from both peripheral and interior junctional sarc...

Phospholamban-dependent effects of C12E8 on calcium transport and molecular dynamics in cardiac sarcoplasmic reticulum.

We have studied the effects of the nonionic detergent C12E8 on Ca-ATPase enzymatic activity and oligomeric state (detected by time-resolved phosphorescence anisotropy, TPA) in skeletal and cardiac sarcoplasmic reticulum (SR). In skeletal, SR, C12E8 inhibits the CA-ATPase, both at high (micromolar and above) and low (submicromolar) Ca. In cardiac SR, C12E8 inhibits at high Ca but activates at lo...

The Effect of pH on the Transient-State Kinetics of Ca^-Mg^-ATPase of Cardiac Sarcoplasmic Reticulum A Comparison with Skeletal Sarcoplasmic Reticulum

The effect of pH on the Ca-Mg-dependent ATPase of sarcoplasmic reticulum (SR) was investigated with a rapid mixing quench-flow apparatus capable of measuring phosphorylation and dephosphorylation at times as rapid as 4 msec. The rates of formation and decomposition of the phosphorylated intermediate (E ~ P) of the Ca-Mg-ATPase were studied in the pH range between 7.6 and 6.0. At pH 6.8, the rat...

Poges 387-394 ACYLPHOSPHATASE STIMULATES Ca 2+ TRANSPORT AND Ca2+-DEPENDENT ATPase ACTIVITY IN CARDIAC SARCOPLASMIC RETICULUM

Acylphosphatase purified from heart muscle actively hydrolyzes the phosphoenzyme intermediate of cardiac sarcoplasmic reticulum Ca2+-ATPase. This effect was evident with acylphosphatase concentrations (up to 100 units/mg sarcoplasmic reticulum .protein) that fall within the physiological range, and the low value of the apparent Kin, on the order of 10t M, suggests a high affinity towards this s...

Effects of phytoestrogens on sarcoplasmic/endoplasmic reticulum calcium ATPase 2a and Ca2+ uptake into cardiac sarcoplasmic reticulum.

Phytoestrogens are naturally occurring estrogenic compounds found in plants and plant products. These compounds are also known to exert cellular effects independent of their interactions with estrogen receptors. We studied the effects of the phytoestrogens phloretin, phloridzin, genistein, and biochanin A on Ca(2+) uptake into the cardiac muscle sarcoplasmic reticulum (SR). Genistein and biocha...
